Smear preparation | ||
1 | Label the Patient form & specimen bottles , Centrifuge tube and slide with labels | CHECK THE PRODUCT |
2 | Mix and shake the sample bottle with hand or shaker until it turns into uniform & cloudy liquid

3 | Took 1 ml from the sample bottle and add 1 ml from the buffer into centrifuge tube | CHECK THE PRODUCT |
4 | Centrifuge, enrich cellular sample and remove non diagnostic impurities for 5 min | |
5 | Remove the filter from the centrifuge tube and throw it then discard the supernatant | |
6 | Insert the slide inside the slide holder and also assemble the settling chamber altogether same as in this picture for | CHECK THE PRODUCT |
7 | Add 0.5 ml to 1 ml from the sample to the settling chamber for 10 – 15 min | |
8 | Add 0.5 ml or 1 ml from the Green Transparent reagent to the settling chamber or you can Rinse the side inside the jar directly for 2-5 min

Smear fixation | ||
9 | Add 1 ml 95% Ethanol 10 minutes (fixation) to the settling chamber or you can Rinse the side inside the jar directly for 10 min | CHECK THE PRODUCT |
Smear staining protocol | ||
1 | Rinse in tap water or the buffer | |
2 | Harris or Gill Hematoxylin 1-3 minutes | |
3 | Rinse in tap water or Scott’s tap water |

4 | 95% Ethanol 10 dips | |
5 | OG-6 – EA-50 stain for 1 – 5 minutes. | |
6 | 95% Ethanol 10 dips, 2 changes | |
7 | 100% Ethanol 1 minute | |
8 | Clear in 2 changes of xylene, 2 minutes each

9 | Mount with permanent mounting medium |
